In a CEM microwave vial with a stir-bar (optional), the nitroaniline (0.36 mmol), carboxylic acid (1.03 mL, 0.35 M), and SnCl2Æ2H2O (3.0 equiv) were combined and heated at 130 C (125 W, 1.0 min ramp time) for 5 min. After cooling, the reaction mixture was diluted with water (4 mL) and neutralized with 50% aq NaOH (pH = 7.0 ± 0.5). It was extracted with ethyl acetate (3 · 4 mL) and the combined organic phases were dried (Na2SO4), filtered and evaporated under reduced pressure. The crude product was analyzed by HPLC/MS/ CLND and purified by passing through a plug of silica gel (500 mg prepacked columns) using 30 mL of ethyl acetate. Compounds with purity below 95% were further purified using the ISCO CombiFlash 16sqx with a gradient of either 5–100% EtOAc–hexanes or 0–20% DCM– MeOH (5, 13, and 21). |

在带有搅拌子的CEM 微波小瓶中，加入 硝基苯胺：0.36mmol 羧酸 ：，0.35M ，1.03ml SnCl2 ：? 水 ：3 当量 在130度加热5分钟后（125W），冷却，混合物用4ml水稀释，并用50%的氢氧化钠溶液中和至pH = 7.0 ± 0.5. 。。。。。。（同楼上） 用硅胶纯化，洗提液是30mL乙酸乙酯 如果该化合物的纯度低于95%，那么要用ISCO CombiFlash 16sqx 进一步纯化， 可以用是5~100%乙酸乙酯-己烷或是0-20%二氯甲烷-甲醇进行梯度洗脱。 |
