Compared with icotinib, the disappearance of the doublet of doublets in the 1H NMR spectrum demonstrated the hydroxylation at C4 or C6. From the HMBC spectrum of M1, the hydroxyl-ation at C4 was identified because the signal disappearance of H4 coupled with the C7 (¦Ä 83.73 ppm). All chemical shifts were expressed in ppm relative to the internal standard . The data for COSY, HSQC and HMBC spectra were acquired with 1024 data points for F2 increments and 256 for F1 increments.
