The enzymatic reactions were carried out at 25℃ in 20 mM phosphate buffer (pH 6.5) containing 1% (w/v) casein and an enzyme level of 20 units per gram of protein substrate. The reaction mixtures were incubated for various times (0, 10, 20, 30, 60 min, respectively), then stopped by directly mixing with the sample buffer (2°) of electrophoresis. The reaction mixtures were analyzed by SDS–PAGE and by measuring the ammonia released (Ngo,Phan, & Yam, 1982). |

酶的反应条件是25度，含有1%的casein（干酪素, 酪蛋白）的PH=6.5的浓度为20mM磷酸缓冲液，每克蛋白的底物是20u。反应体系分别用不同的时间（0, 10, 20, 30, 60 分钟培养），然后终止反应，样品用上样缓冲液（2x）混合后电泳检测。 反应体系样品用SDS-PAGE和采用测定溶液中氨的含量的方法（定氮法？？？）(Ngo,Phan, & Yam, 1982) 金币不要了，就是说的一个反应条件 [ Last edited by sutao1979 on 2009-3-9 at 18:44 ] |
