The effect of Wood-pulp derived sterols on Cholesterol metabolism gene regulation in Hamsters fed High-cholesterol diets
Abstract: Plant sterols, similar to cholesterol, are rich in vegetable oils and products and can lower blood cholesterol, which may reduce the risk of coronary heart disease. However, the mechanism of plant sterols lowering blood cholesterol is still elusive. In this study, we used hamsters fed high cholesterol diets to investigate the effect of different concentration of sterols on cholesterol metabolism. The results suggest that the corn-derived sterol can reduce the weight of liver and the level of TC, TG, HDL-C (p<0.01), significantly. Moreover, the mRNA level of CYP7A1, HMG-CoA-R, LDL-R is up-regulated (p<0.05), which promotes the rate of cholestrol metabolism in liver. The result of gas chromatography demonstrates that corn-derived sterol can reduce the level of cholesterol in liver and increase the excretion of cholesterol in feces. We also investigate the component of microorganism in intestinal tract and find that corn-derived sterol can alter the distribution of microorganism and increase the percentage of probiotics. These results indicate that corn-derived sterol can reduce levels of cholesterol in blood and liver through up-regulating the expression level of cholesterol metabolism genes, preventing the absorption of cholesterol and improving the diversity of microorganisms in intestines.
