关于荧光猝灭HSA的,求助怎么答复审稿人

The equation used by the authors to calculate the "affinity constant Ka" is modified Stern-Volmer (equation (2)), NOT the Scatchard equation. This is completely wrong. Because the modified Stern-Volmer just show the differing accessibilities of tryptophan residues in HSA that resulted in the frequent use of quenching to resolve the accessible and inaccessible residues. Or rather, this plot provides a useful but arbitrary resolution of two assumed classes of tryptophan residues. Except for this point, this relationship has no physical sense. Thus all the discussions based on the modified Stern-Volmer must be deleted in this manuscript. 谢谢。 |
