Recently, protein engineering in combination with in silico methods gave rise to the development of a tailor-made HNL for large-scale manufacturing of a specific target cyanohydrin. Improved evolutionary techniques and efficient screening systems are important prerequisites for straightforward directed evolution programs.Structure/function considerations can be addressed by structural biology and molecular modeling possibly in connection with bioinformatics processing sequence data—which are key elements of successful enzyme development by means of rational design to obtain specifically tailored enzymes. With this respect, the existence of a platform of numerous different—if possible R- and S-selective—enzymes for a given chemical transformation as an “off-the-shelf” technology can be of great value for the fast evaluation of the feasibility of an enzymatic reaction in a multistep synthesis route. This is especially important for early-stage drug development where strict timelines usually impede the laborious and hence time-consuming process of biocatalyst discovery and optimization. |
