SDS-PAGE凝胶制备属于实验室最常规的操作了，刚开始做蛋白实验总是在找凝胶配制实验中所用试剂的配方，这里总结了分离胶、浓缩胶、分离胶和浓缩胶缓冲液、考马斯亮蓝染液、样品缓冲液、电泳缓冲液等配方。 1. 分离胶(12%) 配制 所需试剂 所需体积 分离胶(12％) 30％丙烯酰胺 6.0 ml 1.5 mol/l Tris-HCl 3.8 ml 10%SDS 150 μl 10%过硫酸铵 150 μl TEMED 6 μl 蒸馏水 4.9 ml 2. 浓缩胶(5%) 配制 所需试剂 所需体积 浓缩胶(5％) 30％丙烯酰胺 1.3 ml 1.5 mol/l Tris-HCl 1.0 ml 10%SDS 80 μl 10%过硫酸铵 80 μl TEMED 8 μl 蒸馏水 5.5 ml 3. 分离胶缓冲液(pH8.9) 配制 所需试剂 所需体积 分离胶缓冲液 Tris 36.6 g 1 mol/l HCl 48 ml 蒸馏水 补足至100 ml 4. 浓缩胶缓冲液(pH6.7) 配制 所需试剂 所需体积 浓缩胶缓冲液（pH6.7） Tris 5.98 g 1 mol/l HCl 48 ml 蒸馏水 补足至100 ml 5. 30%丙烯酰胺(pH≤7.0) 配制 所需试剂 所需体积 30%丙烯酰胺（pH≤7.0） 丙烯酰胺 29 g 甲叉-双丙烯酰胺 1 g 蒸馏水 补足至100 ml 6. 10%过硫酸铵(W/V)：1g过硫酸铵溶于10ml水中，4℃保存数周，尽量现用现配。 7. 10%SDS：将10 g SDS溶于80ml重蒸水中并轻缓搅拌(室温低时需水浴加热溶解)，再定容至100ml，室温存放。 8. 样品缓冲液 配制 所需试剂 所需体积 样品缓冲液 1 M Tris-HCl（pH6.7） 5 ml SDS 2.5 g 巯基乙醇 1 ml 溴酚兰 0.05 g 蔗糖 10 g 蒸馏水 补足至100ml 9. 考马斯亮蓝染色液：0.25g考马斯亮蓝R-250用少量甲醇溶解后，用甲醇(40ml)-乙酸(10ml)水溶液稀释至100ml。 10. Tris-甘氨酸电泳缓冲液(pH8.3) 配制 所需试剂 所需体积 Tris-甘氨酸 电泳缓冲液（pH8.3） Tris碱 15.1 g 甘氨酸 94 g 10％（W/V）SDS 50 ml 去离子水 补足至100 ml 凝胶配制所需试剂的母液不能放置太久，有条件的实验室可将母液保存在4度冰箱中，放置母液被微生物污染或者出现沉淀。 |

SDS-PAGE Gel 1. Prepare the separation gel (10%). Mix in the following order: H2O 4.1 mL Acrylamide/bis (30% 37.5:1; Bio-Rad) 3.3 mL Tris–HCl (1.5 M, pH 8.8) 2.5 mL SDS, 10% 100 μL N,N,N′,N′-tetramethylethylene-diamine (TEMED) (Bio-Rad) 10 μL Ammonium persulfate (APS), 10% 32 μL After adding TEMED and APS to the SDS-PAGE separation gel solution, the gel will polymerize quickly, so add these two reagents when ready to pour. 2. Pour gel, leaving ～2 cm below the bottom of the comb for the stacking gel. Make sure to remove bubbles. 3. Layer the top of the gel with isopropanol. This will help to remove bubbles at the top of the gel and will also keep the polymerized gel from drying out. In ～30 min, the gel should be completely polymerized. 4. Remove the isopropanol and wash out the remaining traces of isopropanol with distilled water. 5. Prepare the stacking gel (4%). Mix in the following order: H2O 6.1 mL Acrylamide/bis (30%, 37.5:1) 1.3 mL Tris–HCl (0.5 M, pH 6.8) 2.5 mL SDS, 10% 100 μL TEMED 10 μL Ammonium persulfate (APS), 10% 100 μL 6. Pour stacking gel on top of the separation gel. 7. Add combs to make wells. In ～30 min, the stacking gel should become completely polymerized. 8. Clamp gel into apparatus, and fill both buffer chambers with gel running buffer according to the instructions for the specific apparatus. 9.
