关于固定化酶动力学常数，审稿人的意见请大家提供两篇文献 已有1人参与

论文中测定固定化后Km变大，我用构象的不适改变加以解释。而Vmax的变大，我用了富集效应及减少内扩散来解释。但审稿人的意见是两个结果应该统一用构象改变来解释。以下是审稿人原话：The increased KM and Vmax should be explained in common bases, very likely due to changes in the conformation of the enzyme after immobilization (Rodrigues and Secundo review above study this). Current explanations seem no very likely, why the enzyme activity increase (bit not the Km decerase) using small volume?? Substrate concentration on the support could explain some effect, but the dissimilar effect on Km and V,ax suggest just a conformation change, and very likely, some diffusion limitations, May be interesting repeat the experiment using lower enzyme loading to check if the apparent Km decreased. 还请大家帮帮忙，提供文献支撑。是（Rodrigues and Secundo ）这两个人写的相关文章吗？ |
