首先非常感谢各位浏览我的帖子. 有建设性意见者我发币币哦!!!! xiexei 请问各位,T4,T3 是在水中和在甲醇中的溶解性怎么样? 有人做过组织中甲状腺激素的提取和测定么? 最近看文献有人这样提取大鼠组织中甲状腺激素,这个方法在英文杂志上比较普遍,但是不太明白,请各位帮个忙? 难道甲状腺激素即能溶于甲醇又能溶于水么? 水相通过树脂柱过滤难道能够截流甲状腺激素么? 不好意思,问题有点多,但是主题只有一个   In brief, the frozen weighed tissue aliquot was homogenized directly in methanol. Appropriate volumes of chloroform were added to extract with chloroform-methanol (2:1, vol/vol) twice, followed by back-extraction into an aqueous phase, also twice. The aqueous phases from all back-extractions were pooled and further purified on Bio-Rad AG 1X2 resin columns, from which the iodothyronines were eluted with 70% acetic acid. The eluates containing the labeled tracers were pooled, evaporated to dryness, and extensively counted, either before or after drying, for the calculation of individual recoveries. RIA buffer was then added, and T, and Ta contents were determined by RIAs in triplicate at two dilutions.[ Last edited by moneymaker on 2008-11-13 at 17:22 ] |
