首先非常感谢各位浏览我的帖子. 有建设性意见者我发币币哦!!!! xiexei 请问各位,T4,T3 是在水中和在甲醇中的溶解性怎么样? 有人做过组织中甲状腺激素的提取和测定么? 最近看文献有人这样提取大鼠组织中甲状腺激素,这个方法在英文杂志上比较普遍,但是不太明白,请各位帮个忙? 难道甲状腺激素即能溶于甲醇又能溶于水么? 水相通过树脂柱过滤难道能够截流甲状腺激素么? 不好意思,问题有点多,但是主题只有一个 In brief, the frozen weighed tissue aliquot was homogenized directly in methanol. Appropriate volumes of chloroform were added to extract with chloroform-methanol (2:1, vol/vol) twice, followed by back-extraction into an aqueous phase, also twice. The aqueous phases from all back-extractions were pooled and further purified on Bio-Rad AG 1X2 resin columns, from which the iodothyronines were eluted with 70% acetic acid. The eluates containing the labeled tracers were pooled, evaporated to dryness, and extensively counted, either before or after drying, for the calculation of individual recoveries.
