MBE：所有遗传疾病的基因有同一“祖先”

科学家对很多引起疾病的基因出现的时间进行了系统分析，研究首次发现，这些基因大部分在第一个细胞产生时出现，其它一些基因产生在多细胞有机体首次出现的10亿年前，哺乳动物产生后出现的基因的变异不会引起疾病，研究结果发表在最新一期的《分子生物学和进化》杂志上。 该研究使科学家能更好地认识同遗传疾病有关的基因，而且研究结果也证明，引发疾病的基因间存在相互关系，这也有助于科学家研究一些同多因子疾病相关的基因。 旨在破译人类基因密码的人类基因组计划目前已经揭开了几千个基因的“面纱”，研究发现，如果这些基因发生突变，就会引发人类的遗传疾病。 德国莱比锡马克斯 普朗克进化生物学研究所的托米斯拉夫 多马泽特-罗索和戴萨德 陶特兹使用一种新的统计学方法来分析这些基因，他们通过追踪某一基因的祖先来追溯该基因的源头，接着确定该基因的最小年龄，借此，他们对所有目前存在的基因进行了此类分析。 应用这种方法追踪疾病基因，科学家惊奇地发现：大多数这些疾病基因可以追溯到第一个细胞那里，其它大群的基因出现在10亿年以前———此时，多细胞有机体首次出现。而且，没有疾病同一些哺乳动物产生后出现的基因有关。 这些发现表明，遗传疾病主要影响地球生命最初出现时的细胞过程，据此，我们可以得出结论：现在，地球上所有的生命有机体，不仅是人类，都被同一种遗传疾病影响。而且，这也暗示，遗传疾病不会被人类完全攻克，因为它们同生命最初的进化过程有关。 科学家称，至于为什么进化得越晚的基因，比如同哺乳动物的出现相关的基因发生突变时，不会引起疾病，仍是未解之谜。 |

The majority of these appear to be non-essential genes (i.e. are not embryonically lethal when inactivated) and one could therefore speculate that they are late additions in the evolutionary lineage towards humans. Contrary to this expectation, we find that they are in fact significantly over-represented among the genes that have emerged during the early evolution of the metazoa. Using a phylostratigraphic approach, we have studied the evolutionary emergence of such genes at 19 phylogenetic levels. The majority of disease genes was already present in the eukaryotic ancestor and the second largest number has arisen around the time of evolution of multicellularity. Conversely, genes specific to the mammalian lineage are highly underrepresented. Hence, genes involved in genetic diseases are not simply a random subset of all genes in the genome, but are biased towards ancient genes. |
