本研究对四种表型(全黑、半黑、花斑、全红)萍乡(肉)红鲫(Pingxiang red-transparent crucian carp)的鳞片及皮肤色素细胞进行了光学显微镜观察,并对其皮肤进行了组织学研究,克隆了萍乡(肉)红鲫黑色素浓集激素受体基因的部分片段,并对其在组织中的分布和表达情况进行了研究,同时克隆了萍乡(肉)红鲫的Cyt b基因,分析比较了萍乡(肉)红鲫在进化上的地位。 1.利用光学显微镜对四种表型萍乡(肉)红鲫鳞片及皮肤进行了观察,发现其鳞片都只含有三种色素细胞,即黑色素细胞、黄色素细胞和虹彩细胞;而其皮肤都只含有黑色素细胞和黄色素细胞;不同表型的萍乡(肉)红鲫鳞片和皮肤色素细胞的分布存在差异,主要表现在黑色素细胞的形态和分布上。通过常规石蜡切片技术,对全红萍乡(肉)红鲫的皮肤进行了组织学观察,发现其色素细胞不仅存在于表皮层与真皮层之间、真皮层与皮下层之间,在腺层和皮下层中也有分布; 2.克隆了全红萍乡(肉)红鲫MCHR2基因编码区的部分cDNA序列,长度为366bp,编码122个氨基酸,分子量为14.36 KDa,等电点为10.02;用荧光定量PCR检测了MCHR2基因在全红萍乡(肉)红鲫组织中的分布以及四种表型(肉)红鲫大脑和皮肤组织中的分布,结果发现MCHR2在全红萍乡(肉)红鲫肝脏中表达量最高,在心脏中表达量最低,其次依次是眼、肌肉、大脑、鳃、肾、肠和心脏;在四种表型(肉)红鲫皮肤组织中,MCHR2的表达量无显著差异;而在大脑组织中,全红表型与半黑表型、全黑表型的MCHR2表达量都存在显著差异,而与花斑表型的表达量没有明显差异。 3.克隆了萍乡(肉)红鲫MCHR1a和MCHR1b基因部分cDNA序列,通过BALST比对发现,其间有96%的同源性;利用RT-PCR技术检测发现MCHR1a和MCHR1b在组织中的分布和表达量基本一致,因此,认为萍乡(肉)红鲫MCHR1a和MCHR1b基因是同一个基因,改名为MCHR1,即萍乡(肉)红鲫MCHR1基因并未发生分化,经拼接获得了其编码区的部分cDNA序列,长度为481 bp,编码160个氨基酸,分子量为17.78 KDa,等电点为9.27; 4.克隆了四种表型(肉)红鲫线粒体细胞色素b基因完整编码区,发现其序列完全一致,长度为1 140bp,编码380个氨基酸。表明萍乡(肉)红鲫群体尽管部分个体在表型上存在差异,但并没有在分子水平上发生改变,没有形成亚种。运用邻接法构建的分子系统树表明萍乡肉红鲫与普通鲫鱼亲缘关系最近,而与银鲫的亲缘关系略远。 |
