DNA双链带的负电荷比单链多吗？

做了一个荧光猝灭检测DNA，猝灭剂称为Q。Q的zeta电势为正值，本来解释为Q和荧光标记的ssDNA静电吸引，加入配对DNA之后，形成dsDNA,荧光恢复（文章里面的解释： because of the absence of the unpaired nucleobases and the rigid conformation of the formed P1/T1 duplex……）。但审稿人说形成dsDNA后应该带更多负电荷，说我的解释不合理，实在是没搞明白 审稿意见：However, when the ssDNA (荧光标记的DNA) was hybridized with its complementary strand T1, the dsDNA may show more negative charge. So, here is my question, why dsDNA has weaker interaction with Q from this point. |
