Indeed, when pressure reaches to 18MPa, the particle size of drug will be reduced by 10 times instantaneously. For this situation, we performed additional several repeated tests for verification. The cause for such situation may be due to following two factors. Firstly when pressure is higher than 18MPa, crystal form will be transformed from needle shaped crystal into thin plate shaped crystal. According to literature summary, we can know that the particle size of crystal, which is in form of needle shape during crystallization, is generally larger; Secondly, an increased pressure will lead to an increased degree of supersaturation as well as increased nucleation rate of drug. Regarding Etoposide drug crystals, when pressure reaches 18MPa, it will pass the critical point where the crystal nucleation rate is over crystal growth rate. Therefore, such situation will be caused.
