Substrate micronization So-called “wet” disintegration of steroid particles based on ultrasonic wave in combination with detergents can facilitate the contact between substrate and whole-cell biocatalyst. Organic-aqueous biphasic systems Two-phase organic system based on n-hexane,butanol, benzene etc. may increase the productivity of biotransformation due to dissolution of steroid substrate/product and make easier downstream processing. Major disadvantages concerned inactivation or destruction of biocatalyst in the apolar phase. Water-miscible organic co-solvents Water-miscible organic solvents are used to overcome the low solubility of steroid substrates/products and hence intensify the productivity. Cloud-point systems based on nonionic surfactant This approach was regarded as a novel two-phase partitioning system and used for solubilization and phase separation in whole cell conversion. It enables elimination of substrate and product inhibition, and prevents further degradation of product. Liquid polymer based systems Liquid polymers as components of the bioconversion media (silicon oil, polypropylene glycol, polydimethyl siloxane, etc.) are used for solubilization of sterol compounds and acceleration of steroid accumulation. |
