非常急！！！！悬赏50金币！！求助高手！！ Peptides were separated on a 20-cm column with a 75 µm inner diameter packed with Dr. Maisch GmbH reversed-phase material Reprosil-Pur 120 C18-AQ, 1.9 µm resin (SinoAmerican Proteomics LLC). Separation was performed using 220-min runs on a Waters NanoAcquity liquid chromatography with a binary buffer system of 2% acetonitrile in 0.1% formic acid (buffer A) and 99.9% acetonitrile in 0.1% formic acid (buffer B) interfaced to a LTQ-Orbitrap Velos (Thermo Fisher Scientific). The sample was loaded onto the trap column at a flow rate of 10 µl/min for 1 min and then separated at a flow rate of 300 nl per min by an initial linear gradient of buffer B from 6% to 26% up to 180 min followed by a 26% to 80% buffer B gradient for 40 min. The mass spectrometer was operated in data-dependent mode, with one full scan acquisition in the Orbitrap at 30,000 resolution (at 400 m/z) with an automatic gain control (AGC) target value of 10^6 ions. The 10 most intense ions with a charge >1 were selected for tandem data-dependent analysis using a hybrid method of alternating collision-induced dissociation and higher energy collision dissociation fragmentation techniques (CID and HCD, respectively)1. CID scans were collected at an AGC target value of 5,000 with a maximum injection time of 25 ms. HCD scans were collected at 7,500 resolution with an AGC target value of 30,000 and a maximum injection time of 150 ms. Dynamic exclusion was set to +/- 0.005 m/z for 60 seconds with an early expiration count of 1 and signal to noise of 2.[ 来自科研家族 生化掘金 ] |
