| 本实验通过对四川各地土样的采集，用于ε-多聚赖氨酸(ε-poly-lysine，ε-PL)产生菌种的筛选与鉴定。实验中首先对采集回来的泥土用SG液体培养基进行富集培养48h，再根据ε-PL带正电荷并能与带正电荷的亚甲基蓝排斥形成透明圈的性质，通过加入亚甲基蓝（0.02g/L）和重铬酸钾（0.05g/L，作为抑菌剂）的SG固体培养基对ε-PL产生菌进行初筛。其次，根据ε-PL是一种生物碱，就可以通过改良碘化铋试剂钾（Dragendorff）这种生物碱的显色剂能与DR试剂形成砖红色沉淀和甲基橙的磺酸基阴离子能与聚赖氨酸的氨基阳离子能形成阳离子对，在pH3~10的范围内能形成砖红色沉淀的原理，通过对发酵上清液进行显色复筛。然后，再通过对筛选出的ε-PL产生菌进行生长特征、形态特征、生理生化鉴定初步鉴定筛选出的菌属，再通过16SrDNA的分子生物学鉴定方法最终确定出筛选出的菌种种类。 |
