用ImageJ统计吖啶橙染色结果

各位前辈，小弟最近再做斑马鱼整胚的吖啶橙染色（Acridine Orange staining，AO染色），苦于没有简单高效的统计方法，找到一篇文章是介绍AO染斑马鱼如何用ImageJ统计的方法，可是我看不懂，对IMAGEJ不了解，先摘下来各位大神帮我看看：最好附出来步骤。 Images were manipulated and analyzed using ImageJ software (National Institutes of Health, NIH). The image background surrounding the embryo was removed using the threshold tool, and all embryos in an image were then selected using the wand tool. ImageJ was then used to generate a histogram of pixel density from the selection to report, among other statistics, the median value of pixel density. ----------------- Images were manipulated and analyzed using ImageJ software (National Institutes of Health, NIH). During this procedure, to facilitate measurement of light only from embryo images, a uniform threshold value (30) was set below which background light intensity levels were reduced to zero (black). In the densitometric analysis, background pixels were excluded by using the wand (tracing) tool to select only pixels representing the embryo. The measurement tool was then used to calculate the median pixel value. The histogram tool was used to produce histograms in Figures 1 and 2.  |
