2.8. Measurement of rat platelet adenylate cyclase (AC) and guanylate cyclase (GC) Washed platelet after ultrasonication was centrifuged at 11,000 g for 15 min . AC was into the supernatant and the GC into precipitation. The concentrations of AC and GC were measured and adjusted to 1 mg/ml. Reaction systemof 200 µl included the following composition (mmol/L): GTP 0.2 or ATP 0.5, Tris–HCl 50 (pH 8.0), KCl 200, ATP 0.1, MgCl2 1, DTT 2, PMSF 1, GC or AC 50 µl. Reaction started from the mixing of reaction system at 37 °C for 15 min and concluded by 10% perchloric acid of 300 µl, following the neutralization of 1.01 mol/L KOH of 500 µl and centrifugation at 825 g for 10 min. cAMPand cGMP insupernatants was were measured with a cAMP and cGMP EIA system.
