A polysaccharide ,from which can sepratede P-1 and P-2 by DEAE cellulose-52 chromatography and Sephadex G-100 size-exclusion chromatography, was extracted from herb-A using ultrasonic-assisted extraction. Then identify the Structural characterizations of P-1 and P-2 ,including molecular weight, monosaccharides composition, sulphate and uronic acid contents, conformational structure, ultraviolet spectrum, and infrared spectrum, and study their anti-proliferation activities against human gastric cancer BGC-823 cells and lung cancer A549 cells in vitro. It can be seen from the research that both P-1 and P-2 can significantly inhibit the proliferation of BGC-823 cells in a certain concentration. The highest inhibitory rates were 78% for P-1 and 95% for P-2 at the concentration of 200 μg/ml. This higher anticancer activity may be attribute to the structural characterization of polysaccharides, and the recognition of biological system for the conformational structure of polysaccharides as well . |
