Antibacterial peptides are a kind of polypeptide with antimicrobial activity produced by microorganisms, plants, invertebrates, and various animal cells and tissues, whic h are important component of innate immune system of biosystem in vivo. Due to their distinct antibacterial mechanism of antibiosis, antibacterial peptides are expected to become the next generation of anti-microbial drugs. But the source of antibacterial peptides has always been the bottleneck of their researches and utilizations. In order to solve the problem of their source, this paper successfully expressed antimicrobial peptides LNK-16 with antimicrobial activity with bio-engineering techniques and methods.
At first, with the methods of bioinformation, we found the cattle antibacterial peptide Indolicidin with high antibacterial activity, broad antibacterial spectrum, low molecular weight and linear structure in the database of NCBI, which containing 13 amino acid peptides. In order to achieve the expression of Indolicidin in E.coli, by adding amino acid recognition sites of kallikrein (PFR) at the N terminal,we synthesized the new peptide named LNK-16. The detection of antibacterial activity showed that LNK-16 also had antibacterial activity. |
