The S0-oxidizing bacterial consortium capable of reducing
ClO4 to Cl was enriched from municipal activated sludge.
The culture was enriched in 300 mL serum bottles. A
volume of 6 mL activated sludge sample (5% v/v) was added
to 120 mL of culture medium containing 3 mMperchlorate,
20 mM S0 powder, and basal medium amended with
trace metal solution I and YE. After purging the headspace
with N2/CO2 (80:20, v/v), the enrichment cultures were
incubated in the dark at 308C on a shake table (100 rpm).
When ClO4 was depleted, the culture was transferred again
by inoculating enriched culture (5%, v/v) to fresh medium.
The culture was transferred every 21–25 days.
大家帮忙，看看，我就感觉如果每个富集周期结束以取5%的体积，开始下一个富集周期，感觉不太合理。想问问大家正确理解是这样吗？
注：ClO4——高氯酸根。Cl——氯离子。S0——硫单质。

“When ClO4 was depleted, the culture was transferred again by inoculating enriched culture (5%, v/v) to fresh medium.”

是这样。培养基是120 mL, 5%体积比是 6 mL。最初接种的活性污泥也是 6 mL，进一步富集硫氧化菌时也应该用 6 mL, 也就是5%体积比。
