| The synthesis of both the piperazine and the homopiperazine analogs that are described in Tables - 4 was achieved via amide bond coupling of the corresponding 3-[(4-oxo-3H-phthalazin-1-yl)methyl]benzoic acid 7 or 2-fluoro-5-[(4-oxo-3H-phthalazin-1-yl)methyl]benzoic acid 8 (Scheme 1). The synthesis of corresponding intermediate 7 is outlined in Scheme 1. The nucleophilic addition of 3Hisobenzofuran-1-one 1 to 3-formyl-benzonitrile 2 in the presence of sodium methoxide provided 3-(1,3-dioxoinden-2-yl)benzonitrile 3 in good yield. |

| 表4中所述的哌嗪和高哌嗪同系物的合成都是由相应的3-[(4-oxo-3H-phthalazin-1-yl)methyl]苯甲酸7或2-氟-5-[(4-oxo-3H-phthalazin-1-yl)methyl]苯甲酸8经酰胺键耦合而实现的（见示意图1）。有关的中间体7的合成在示意图1中标明。在甲醇钠存在下，3H-异苯并呋喃-1-酮 1向3-甲酰基苯腈 2的亲核加成生成3-(1,3-dioxoinden-2-yl)苯甲腈的反应具有良好的产率。 |
