5.1 Method Summary
Samples are dissolved in methanol.make methanol +0.05% H3PO4 the mobile phase. ZORABX80AExtend a Cl8 stainless steel column is used. variable wavelength UV detector is set at 260nm wavelength.
Separation and determination of rhein, emodin, chrysophanol in the samples are quantified by an external standard mode.
Emodin is used as standard sample to separate and determine the content of aloe emodin in the parent drug by correction methods.
Determination procedure
About 0.149 (accurate to 0.0002g) and 0.02g standard products are weighed and put in 100mL volumetric flask, diluted with methanol to scale, shake, and inject samples according to the following sequence : a. Standard sample solution, b. Sample solution, c. Sample solution, d. Standard sample solution.
5.5 Calculation
Work out the average peak area of the two sample solution and two-pin standard samples before and after of rhein, emodin, chrysophanol and physcion,respectively.
Contents in x (%) of aloe emodin, rhein, emodin, chrysophanol and physcion in samples are calculated as follows:
PS:
Al means the average peak area of an adjacent two-pin standard sample solution of rhein, emodin, chrysophanol and Physcion ;
A2 means the average peak area of adjacent two-pin sample solution of aloe emodin, rhein, emodin, and emodin chrysophanol ;
m1 1 means quality of the rhein, emodin, chrysophanol and Physcion standard sample, g; m2: the quality of the sample, g;
P means the wt% of rhein, emodin, chrysophanol and physcion in the sample. |
