AB Microemulsion liquid chromatography (MELC) is defined as a kind of liquid chromatography using microemulsions as mobile phases, in which water-in-oil and oil-in-water microemulsions are used on normal and reverse phase columns, respectively, while the detectors are the same as conventional liquid chromatography. Many specific advantages of MELC have been demonstrated, including unique selectivity, high efficiency, the ability to separate simultaneously compounds covering diverse solubility and to inject directly plasma samples onto the HPLC system and elimination of the need for an equilibration rinse between gradients. This article introduced the composition and physicochemical structures of different microemulsions used in MELC, and discussed the effects of operating parameters on separation in detail. Some applications were illustrated and a promising outlook of MELC was described.
