Wainer. -- 2nd ed., rev. and expanded. page：443 ISBN 0-8247-8819-2 (allc. paper) 本书第一版就预示了药物立体构象在实际应用中的重要性，在过去的几年里，这一推测成为了现实。立体化学也就顺理成章的成为了几乎所有药学科学研究的主题，这方面出现了大量的书籍杂志：《natural andb iological science》《Chirality》 and 《Tetrahedron Asymmetry》相关的学术会议和高校课程更是雨后春笋般出现。目前欧洲，北美，日本的药物管理机构在药物立体异构的药理毒理研究相对更多，成果当然也就多了（希望国内的虫友们多多努力啊，早日摆脱“仿制专家”的“荣誉称号”哦，加油！！！！） 而第二版（即本书更是将立体化学作为药物研发和药理分析的重点，其中4~~6章对立体异构的层析技术做了大量的阐述，第八章，讲述了是药物的酶催化异构体转化合成，第九章讲述药物立体异构体的毒理以及生物转化。第十，十一描述了立体转化以及生物利用度和生物等效性。第十二章则讲述了蛋白质中的立体结合，十三到十六章则重点介绍立体异构的药物的调整，工业生产，以及手性问题。 PREFACE TO THE SECONDED ITION. The first edition of this book contained the observation that the stereoisomeric compositioonf drug substancesw as rapidly becoming a key issue in the development, approvaln, d clinical useo f pharmaceuticals. During the past four years, this assertion has become a reaanlidty s, tereochemistry is now a major theme in all branches of pharmaceutical science. The interest in this topic is reflected in the content of articles appearing in natural andb iological science journaalns d two new international journals, Chirality and Tetrahedron Asymmetry. Moreover, a variety of conferences and courses have been devoted to this subjecInt. addition, the European, North American, and Japanese drug regulatory agencies have expressed an interest in a closer examination of the stereoisomeric composition of chiral drugs and the therapeutic and toxicological consequences of this composition. The pharmaceuticailn dustry has quickly respondedto this situation by considering stereochemistry in its initial drug evaluation strategies. In fact, at the present time, a numofb ecro mpanies have made the decision to market only single-isomer drugs. In lighto f these developments,w e felt that iwt as necessary to review the contenot f the first edition aton du pdate or supplement the information presented in this work. The new topics examineidn this edition include: (1) enzymatic synthesis and resolution of enantiomerically pure compounds (Chapter 8); (2) toxicological consequences and implications of stereoselective biotransformations (Chapter 9); (3) stereoselective transport across epithelia (Chapter 10)(;4 ) anssde ssment of bioavailabilitya nd bioequivalence of stereoisomeric drugs (Chapter 11).,The chapter on stereoselective protein binding (Chapter 12) has been completely rewrainttedn new contributions are presented on the regulatory, induasntrdia cl,l inical aspects of stereoisomeric drugs (Chapters 13-16). In addition, the chapters Ix X Preface to the Second Edition discussing stereoselective chromatographic separations (Chapters 4-6) have been revised and expanded.
