To determine if the as-proposed method can be applied in real samples, the spiked human serum samples at three different concentrations of MTase (0.1, 1 and 10 U/mL) were analyzed using the proposed sensing assay. Here the human serum sample is without MTase (diluted 10 times with 50 mM Tris-HCl buffer) samples. The responses of the signal for Dam MTase in diluted human serum samples were compared with those obtained from Dam MTase in buffer solution.Moreover, three independent spiked serum samples (0.1, 1 and 10 U/mL) were tested by the proposed sensing assay, and the activity of Dam MTase in the spiked serum was calculated by using the calibration curve of standard Dam MTase The recoveries of three independent spiked serum samples were found to be in the range of 95.2%-102.4%, respectively. The precision of the sensing assay was evaluated. The RSDs were in the range of 2.7%-4.5%. These results indicated the potentiality of the proposed method for detection of DNA MTase in real biological samples. 是这一段，也没看出来什么问题呢。 |

你自己稿子里写的serum sample里是没有MTase的。那么实际的serum sample里到底是有没有MTase的。你测这个实际样品的目的是为了考察能不能在serum sample里测你的MTase，那如果serum里本来就没有MTase，你测实际样品的意义在哪里。 你那个without用的就很诡异，你怎么知道without，你用什么方法测过就说without？你应该用标准方法（国标等）去测，测不出来也不能说without，应该说ND (not detected，未检出)。审稿人是想知道MTase在你这个实际样品里的一般浓度是多少，随便打个比方比如说谷丙转氨酶正常人是5-40每毫升。高于5或者低于40就不正常。那你的方法就应该覆盖这个范围，同时要前后更宽一点，才算是可以用的方法。如果serum正常里面是没有的，那你就说通过什么仪器检测了，未检出。但其实为了数据更充分你都应该加一个serum里面本来就有用你的方法可检出的量的MTase的样品（不是spiked的）一起比较，这样才最有说服力。 |
