询问一下这个REVIEWER COMMENTS 接受的概率？？？敬请各位赐教！ 已有2人参与

前几天收到的major revision的修回意见，感觉修改集中在： 1，数据处理；这个只怕需要好好解释解释 2，文字修改，中间小问题不少，其中提出来重写conclusion 的。 3，有一个提了WB的实验问题（做多基因表达方面的，我构建多个表达载体后只是核酸、蛋白质电泳验证了一下，然后测了酶活，对比比较下游工程菌性能），但是不确定是否要补做还是解释解释就行。材料已经很久了，确实补做起来难度不小，45天根本不行。 疑虑： 1，从表面的COMMENTS来说，貌似数据、文字修改方面的建议更多，但不知为啥是个major revision(个人理解补充实验才是这个结论)，是不是因为这方面问题很大还是？？ 2，COMMENTS中提到了WB实验，是否一定要补充实验？ 3，大家评评接受几率有几何？？？？ 4, 这种情况45天的期限，啥时候修回最合适？ 附注意见： ================================================= REVIEWER COMMENTS FOR THE AUTHOR: Reviewer #1: Page numbering needs to be done. The manuscript needs editing; the research findings by authors are not well communicated. Under materials and methods: 1) Plasmid construction and identification, please specify the source of each gene p, m and x. 2) Enzyme assays, line 63, it should be just 1.0mM IPTG and not per L. Also supplementary table 1 says, 'constitutively expressed' and the authors are adding IPTG for enzyme expression, please use correct terminology. 3) Same para provide details of procedures, not just references. While defining unit (line 66 to 68), please provide enzymatically catalyzed reaction's product name and not the enzyme name by itself. 4) Same page please specify 1:10 bath ratio (line 72). Did the authors use equal no of cells of the two strains used in bacterial degumming of ramie fibers? Under results and discussion: Recombinant construction and identification Fig.2: Make it supplementary fig. So now fig. 3 becomes fig.2. Please tabulate the data on enzyme name, resp. gene size and protein molecular weight obtained ( Insert as Table 1). The results provided by authors , line 89-91, doesnot make sense. Enzyme assays : Cannot be a title Supplementary table 2 move to the text, so now becomes Table 2; fig.4 becomes fig.3...and so on. 193% to 2,969% (line 100) doesnot make sense. Lines 104 to 108: Authors properly discuss the relevance of work carried out by the two different groups with your work. Bacterial degumming: Line 113, how authors reached to those percentage values? Line 116, provide reference. Line 119, again how the percentage values were determined? Line 124 to 127: Extracellular enzyme secretion by E coli is the issue here and not any undiscovered factor. Re-write conclusions, seek help of professional manuscript writing agency if needed. Fig.3 (original), now fig.2, Any antibodies available? Any Western performed to confirm recombinant protein? Supplementary Table 3, label as figure. Supplementary Table 4 (original), now Table 2: please provide control activity of pET/BL21 for all the 3 enzymes. Also need to express enzyme activity per g cell dry mass and not U/ml. Supplementary Table 5, What is the significance of CK? Figure Captions: Fig.4: 1,2,3 please specify neatly. If 1 is pel-xyl-man, fig shows pel expresses better irrespective of xyl and no synergism |

1，从表面的COMMENTS来说，貌似数据、文字修改方面的建议更多，但不知为啥是个major revision(个人理解补充实验才是这个结论)，是不是因为这方面问题很大还是？？ 可能是修改的量比较大所致。 2，COMMENTS中提到了WB实验，是否一定要补充实验？ 时间不够可以申请延期。如果没有条件补充实验，最好能找到比较权威的文献说明。 3，大家评评接受几率有几何？？？？ 要看楼主的修改是否到位，回答能否令审稿人满意。 4, 这种情况45天的期限，啥时候修回最合适？ 只要修改到位就可以修回，不用刻意拖延。 |
