Acotiamide Hydrochloride Hydrate (Acofide®  Research code: Z-338 in Zeria; YM-443 in Astellas 1 General Information ● Acotiamide is a peripheral acetylcholinesterase (AChE) inhibitor and the first-in-class drug to treat functional dyspepsia (FD) in the world, which was approved in 2013 by PMDA of Japan. ● Acotiamide was originally discovered by Zeria, and co-developed with Astellas. The drug is co-marketing in Japan. ● Acotiamide prevents the hydrolysis of acetylcholine by inhibiting peripheral AChE which results in the enhancement of impaired gastric motility and consequently the improvement of FD symptoms. ● Acotiamide is indicated for the treatment of postprandial fullness, upper abdominal bloating, and early satiation due to FD. ● Available as tablets with each containing 100 mg of acotiamide and the recommended starting dose is one tablet three times daily before meals.
