甘露醇TLC鉴别显色问题

请教各位，现在按照欧洲药典检测甘露醇的TLC鉴别问题，按照药典操作后不显色， 欧洲药典操作步骤如下：TLC plate: Use ready made TLC plate coated with silica gel G. 薄层色谱板：用硅胶制成的TLC薄层 Preparation of mobile phase: Prepare a mixture of 10 volumes of water, 20 volumes of ethyl acetate and 70 volumes of propanol. 流动相的制备：水：乙酸乙酯：异丙醇（10:20:70），混合均匀。 Reference solution (a): Dissolve 12.5 mg of Mannitol CRS/standard in water and dilute to 5 mL with the same solvent. 对照品溶液（a）：在水中溶解12.5毫克甘露醇标准品，用相同溶剂稀释至5毫升 Reference solution (b): Dissolve 12.5 mg of Mannitol standard and 12.5 mg of Sorbitol standard in water and dilute to 5 mL with the same solvent. 对照品溶液（b）：在水中溶解12.5毫克甘露醇标准品和12.5毫克山梨醇标准品，用相同溶剂稀释至5毫升。 Test solution: Take 25 mg of substance to be examined in water and dilute to 10 mL with the same. 测试溶液：取25毫克的物质，在水中进行检查，用相同溶剂稀释至10毫升。 4-Aminobenzoic acid solution: Dissolve 1 g of 4- aminobenzoic acid in a mixture of 18 mL of anhydrous acetic acid (Water content of anhydrous acetic acid NMT 0.4%), 20 mL of water and 1 mL of orthophosphoric acid. Immediately before use, mix 2 volumes of the solution with 3 volumes of acetone. 对氨基苯甲酸溶液：将1克对氨基苯甲酸溶解在18毫升无水乙酸（无水乙酸含水量不超过0.4%），20毫升水和1毫升磷酸的混合溶液中。使用前立即将2体积的溶液与3体积丙酮混合在一起。 Procedure: Apply 2 uL of each of the solutions on the TLC plate and dip in the mobile phase chamber. Develop over a path of 2/3rd of the plate and remove the plate from chamber. Allow the plate to dry in air. Spray with freshly mixed 2 volume of 4-aminobenzoic acid solution and 3 volume of acetone solution. Dry the plate in a current of cold air until the acetone is removed. Heat the plate at 100 °C for 15 minutes. Allow to cool and spray with a 2% w/v solution of sodium periodate. Dry the plate in a current of cold air. Heat the plate at 100 °C for 15 minutes. Allow to cool and record the observations. The test is not valid until the reference solution (b) shows two clearly separated spots. 程序：将各个溶液分别吸取2ul点样在TLC薄层板，TLC 薄层板倾斜放入在展开缸中，当展开剂超过薄层板的2/3时，将薄层板从展开缸中拿出，让薄层板在空气中干燥。新鲜混合2体积对氨基苯甲酸溶液和3体积丙酮溶液。在冷空气中干燥，直到除去丙酮后，在100°C加热薄层板15分钟。冷却后用2% w/v的高碘酸钠溶液喷雾，在冷空气中干燥薄层板，在100°C加热板15分钟。放冷后观察记录现象。该测试是无效的，直到参考溶液（b）显示两个明显分离的斑点。 我现在做的图片如下，大家有类似情况吗？  picture 2.jpg |
